Methodology
Manual—Microscopic Examination of Cells
Includes segmented neutrophils, neutrophilic bands, lymphocytes, monocytes, eosinophils, basophils, erythrocyte morphology, and platelets.
Specimen Requirements
Whole Blood
Specimen must arrive within 24 hours of draw.
Smear preparation on specimens older that 4 hours may exhibit a loss of cellular integrity.
Draw blood in a lavender-top (K2-EDTA) tube(s). For pediatric specimens, draw blood in a lavender-top (K2- EDTA) BD MICROTAINER® MAP microtube. Label specimen with patient’s name (first and last), date of birth, date and time of collection and the phlebotomist’s initials. Send whole blood refrigerated. Do not freeze.
*Collection tube must be filled to ½ of its intended total volume to ensure adequate blood to anticoagulant ratio.
Specimen stability: 24 hours at room temperature or kept refrigerated at 2-8°C
Minimum volume: 250 µL
Specimen rejection criteria:
Clotted blood
Samples that are > 24 hours old
Collection tubes that are less that one-half the intended total volume
